In old english, nominative/accusative/genitive singular “brōþor” and dative singular “brēþer”.

brōþor -> brother
brēþer + -en -> brethren (common in middle english)
brōþor + -s -> brothers (rare in middle english)

(þ is pronounced as modern th)

Just a note.

“Brethren” is already plural, no need for an “S” at the end. I believe it’s an old plural form, similar to “children” and “oxen”, two of the few remaining “-en” plural forms in the English language.
